Myloi belongs to the Municipal Unit of Pythagoreio within the Municipality of Eastern Samos. According to the 2011 census, the village had 249 residents.

Myloi is considered one of the oldest villages on the island. Its name (โMyloiโ meaning โmillsโ in Greek) is believed to originate from the old watermills that once operated along the Imvrasos River, which powered local agriculture and daily life.
